PPO-Inhibiting Herbicides and Structurally Relevant Schiff Bases: Evaluation of Inhibitory Activities against Human Protoporphyrinogen Oxidase

چکیده
The study of human protoporphyrinogen oxidase (hPPO) inhibition can contribute significantly to a better understanding some pathogeneses (e.g., porphyria, herbicide exposure) and the development anticancer agents. Therefore, we prepared new potential inhibitors with Schiff base structural motifs (2-hydroxybenzaldehyde-based bases 9–13 chromanone derivatives 17–19) as structurally relevant PPO herbicides. inhibitory activities (represented by half maximal concentration (IC50) values) enzymatic interactions hPPO melting temperatures) these synthetic compounds commercial herbicides used against were studied protoporphyrin IX fluorescence assay. In case herbicides, significant changes in temperature observed for oxyfluorten, oxadiazon, lactofen, butafenacil, saflufenacil, oxadiargyl, chlornitrofen, especially fomesafen. Nevertheless, did not display activity or temperature. However, designed model based on determined IC50 values docking (by using AutoDock) found important parts motif inhibition. This could be predict herbicidal toxicity improve design inhibitors.

A codon deletion confers resistance to herbicides inhibiting protoporphyrinogen oxidase.
Herbicides that act by inhibiting protoporphyrinogen oxidase (PPO) are widely used to control weeds in a variety of crops. The first weed to evolve resistance to PPO-inhibiting herbicides was Amaranthus tuberculatus, a problematic weed in the midwestern United States that previously had evolved multiple resistances to herbicides inhibiting two other target sites. Evaluation of a PPO-inhibitor-r...

15 صفحه اولSynthesis and Biological Evaluation of New Thiosemicarbazone Derivative Schiff Bases as Monoamine Oxidase Inhibitory Agents.
Twenty-six novel thiosemicarbazone derivative B1-B26 were synthesized via condensation reactions between the corresponding thiosemicarbazides and aldehydes. The chemical characterization of the compounds was carried out by infrared (IR), mass (MS), proton and carbon nuclear magnetic resonance (¹H- and 13C-NMR) spectroscopic analyses. The compounds were investigated for their monoamine oxidase A...
متن کامل1(Heterocyclyl),2,4,5-Tetrasubstituted Benzenes as Protoporphyrinogen-IX Oxidase Inhibiting Herbicides
It is well known that agrochemicals have played a important role in agricultural production that provided for about 700 million people during the past 50 years. At the same time, the increasing world population seems to be a major driving force for the need to enhance the output of food production. The agrochemical industry has been very successful in developing new herbicides and other agroche...
متن کاملProtoporphyrinogen oxidase as a molecular target for diphenyl ether herbicides.
Diphenyl ether herbicides induce an accumulation of protoporphyrin IX in plant tissues. By analogy to human porphyria, the accumulation could be attributed to decreased (Mg or Fe)-chelatase or protoporphyrinogen oxidase activities. Possible effects of acifluorfen-methyl on these enzymes were investigated in isolated corn (maize, Zea mays) etioplasts, potato (Solanum tuberosum) and mouse mitocho...
